Poldip2通过调控黏附分子表达影响炎症所致的脑内皮细胞损伤

目的 探讨Poldip2在炎症所致的脑内皮细胞损伤中的作用及机制.方法 通过细胞转染建立沉默Poldip2的脑微血管内皮细胞系bEnd.3,ELISA方法检测上清液中炎症因子IL-1β和TNF-α的水平;Western blot方法检测沉默Poldip2对血脑屏障相关蛋白ZO-1和Claudin-5的表达以及黏附相关蛋白ICAM-1及VCAM-1的影响.结果 Poldip2沉默可以减轻LPS诱导的脑内皮细胞炎症因子IL-1β和TNF-α的分泌,恢复血脑屏障相关蛋白ZO-1和Claudin-5的表达,减少白细胞黏附分子ICAM-1及VCAM-1的表达(P<0.05).结论 Poldip2可以通过促进白细胞黏附相关蛋白的表达,造成脑内皮细胞损伤,影响血脑屏障的完整性.
Poldip2 regulates inflammation-induced brain endothelial cell injury by regulating adhesion molecules
Objective To investigate the mechanism of Poldip2 in inflammation-induced brain endothelial cell injury.Methods Establishing siPoldip2 bEnd.3 cell line through cell transfection.The levels of inflammatory cytokines IL-1β and TNF-α in the supernatant were detected by ELISA.The effects of silencing Poldip2 on the expressions of blood-brain barrier related proteins ZO-1 and Claudin-5,and adhesion related proteins ICAM-1 and VCAM-1 were detected by Western blot.Result Poldip2 silence could reduce the secretion of LPS-induced inflammatory cytokines IL-1 β and TNF-α in brain endothelial cells,restore the expressions of blood-brain barrier related proteins ZO-1 and Claudin-5,and reduce the expressions of white blood cell adhesion molecules ICAM-1 and VCAM-1(P<0.05).Conclusion Poldip2 could promote the expression of leukocyte adhesion related proteins,which caused damage to brain endothelial cells,and affect the integrity of the blood-brain barrier.
