Objective To investigate the mechanism of Poldip2 in inflammation-induced brain endothelial cell injury.Methods Establishing siPoldip2 bEnd.3 cell line through cell transfection.The levels of inflammatory cytokines IL-1β and TNF-α in the supernatant were detected by ELISA.The effects of silencing Poldip2 on the expressions of blood-brain barrier related proteins ZO-1 and Claudin-5,and adhesion related proteins ICAM-1 and VCAM-1 were detected by Western blot.Result Poldip2 silence could reduce the secretion of LPS-induced inflammatory cytokines IL-1 β and TNF-α in brain endothelial cells,restore the expressions of blood-brain barrier related proteins ZO-1 and Claudin-5,and reduce the expressions of white blood cell adhesion molecules ICAM-1 and VCAM-1(P<0.05).Conclusion Poldip2 could promote the expression of leukocyte adhesion related proteins,which caused damage to brain endothelial cells,and affect the integrity of the blood-brain barrier.
