目的 探讨黄芩苷对前列腺癌细胞LNCaP增殖和侵袭的影响及可能机制.方法 将前列腺癌细胞LNCaP分为对照组、黄芩苷组和黄芩苷+si-miR-145-5p组,qRT-PCR检测各组细胞中miR-145-5p的表达,MTT法、Transwell实验分别检测细胞增殖、侵袭能力.结果 与对照组相比,黄芩苷组LNCaP细胞增殖能力和侵袭能力下降(P<0.05),细胞中miR-145-5p表达升高(P<0.05);与黄芩苷+miR-NC组相比,黄芩苷+si-miR-145-5p组LN-CaP细胞增殖能力和侵袭能力升高(P<0.05),细胞中miR-145-5p表达降低(P<0.05).结论 黄芩苷可通过上调miR-145-5p抑制前列腺癌细胞LNCaP增殖和侵袭.
Baicalin inhibits the proliferation and invasion of prostate cancer cell LNCaP by up-regulating miR-145-5p
Objective To explore the effect and mechanism of baicalin on the proliferation and invasion of prostate cancer cell line LNCaP.Methods Prostate cancer LNCaP cells were divided into control group,baicalin group,and baicalin+si-miR-145-5p group.The expression of miR-145-5p in each group of cells was detected by qRT-PCR,and the proliferation and invasion abilities of the cells were measured by MTT and Transwell assay.Results Compared with the control group,the ability of proliferation and invasion of LNCaP cells in the baicalin group were decreased(P<0.05),and the expression of miR-145-5p in the cells was increased(P<0.05).Compared with baicalin+miR-NC group,the ability of proliferation and invasion of LNCaP cells in baicalin+si-miR-145-5p group were increased(P<0.05),and the expression of miR-145-5p in cells was decreased(P<0.05).Conclusion Baicalin can inhibit the proliferation and invasion of prostate cancer LNCaP cells by up-regulating the expression of miR-145-5p.
