灯盏花乙素调节SDF-1/CXCR4轴保护脓毒症大鼠心肌损伤

目的 探讨灯盏花乙素调节SDF-1/CXCR4信号通路对脓毒症大鼠心肌损伤的影响.方法 将SD大鼠分为假手术组、模型组、低剂量灯盏花乙素组、高剂量灯盏花乙素组、高剂量灯盏花乙素+AMD3100(CXCR4抑制剂)组,每组12只.采用盲肠结扎穿孔法制备大鼠脓毒症模型;血液生化分析仪检测心肌损伤标志物血清肌酸激酶(CK)、乳酸脱氢酶(LDH)、心肌肌钙蛋白(cTnⅠ)水平;ELISA法检测大鼠血清IL-6、IL-1β、TNF-α水平;TUNEL染色检测心肌细胞凋亡情况;HE染色观察大鼠心肌组织病理学变化;商品化试剂盒检测GSH、SOD、MDA水平;Western blot检测大鼠心肌组织中Bcl-2、Bax、SDF-1、CXCR4蛋白表达.结果 与假手术组相比,模型组大鼠心肌组织有严重损伤,血清CK、LDH、cTnⅠ、IL-6、IL-1β和TNF-α水平、心肌细胞凋亡率、心肌组织MDA水平、Bax蛋白水平显著升高,心肌组织GSH、SOD水平、Bcl-2、SDF-1、CXCR4蛋白水平显著降低(P<0.05);与模型组相比,低、高剂量灯盏花乙素组大鼠心肌组织损伤显著减轻,血清CK、LDH、cTnⅠ、IL-6、IL-1β和TNF-α水平、心肌细胞凋亡率、心肌组织MDA水平、Bax蛋白水平显著降低,心肌组织GSH、SOD水平、Bcl-2、SDF-1、CXCR4蛋白水平显著升高(P<0.05);与高剂量灯盏花乙素组相比,高剂量灯盏花乙素+AMD3100组大鼠心肌组织损伤显著加重,血清CK、LDH、cTnⅠ、IL-6、IL-1 β和TNF-α水平、心肌细胞凋亡率、心肌组织MDA水平、Bax蛋白水平显著升高,心肌组织GSH、SOD水平、Bcl-2、SDF-1、CXCR4蛋白水平显著降低(P<0.05).结论 灯盏花乙素通过激活SDF-1/CXCR4信号通路可降低脓毒症大鼠炎症反应和氧化应激,减轻脓毒症大鼠心肌损伤.
Scutellarin regulates SDF-1/CXCR4 axis to protect myocardial injury in sepsis rats
Objective To investigate the effect of Scutellarin on myocardial injury in septic rats by regulat-ing the SDF-1/CXCR4 signaling pathway.Methods SD rats were separated into sham surgery group,model group,low-dose Scutellarin group,high-dose Scutellarin group,high-dose Scutellarin+AMD3100(CXCR4 inhibitor)group with 12 rats in each group.The cecal ligation and perforation method was applied to prepare a rat sepsis model;blood biochemical analyzer was applied to detect serum creatine kinase(CK),lactate dehydrogenase(LDH)and myocardial troponin Ⅰ(cTnⅠ)levels;ELISA method was applied to detect the levels of IL-6,IL-1β,and TNF-α in serum;TUNEL staining was applied to detect cardiomyocyte apoptosis;HE staining was applied to observe patholog-ical changes in myocardial tissue;commercialized reagent kits were applied to detect levels of GSH,SOD,and MDA;Western blot was applied to detect the expression of Bcl-2,Bax,SDF-1,and CXCR4 proteins in myocardial tissue.Results Compared with the sham surgery group,the myocardial tissue in the model group was severely dam-aged,the levels of serum CK,LDH,cTnⅠ,IL-6,IL-1β and TNF-α,myocardial cell apoptosis rate,the level of myocardial tissue MDA,and the protein level of Bax increased obviously,the levels of GSH,SOD,the protein lev-els of Bcl-2,SDF-1,and CXCR4 in myocardial tissue reduced obviously(P<0.05);compared with the model group,the myocardial tissue damage of rats in the low and high dose Scutellarin groups was obviously reduced,the levels of serum CK,LDH,cTnⅠ,IL-6,IL-1β and TNF-α,myocardial cell apoptosis rate,the level of myocardial tissue MDA,and the protein level of Bax reduced obviously,the levels of GSH,SOD,the protein levels of Bcl-2,SDF-1,and CXCR4 in myocardial tissue increased obviously(P<0.05);compared with the high-dose Scutellarin group,the myocardial tissue damage of rats in the high-dose Scutellarin+AMD3100 group was obviously increased,the levels of serum CK,LDH,cTnⅠ,IL-6,IL-1β and TNF-α,myocardial cell apoptosis rate,the level of myocardi-al tissue MDA,and the protein level of Bax increased obviously,the levels of GSH,SOD,the protein levels of Bcl-2,SDF-1,and CXCR4 in myocardial tissue reduced obviously(P<0.05).Conclusion Scutellarin can reduce inflam-matory response and oxidative stress in septic rats by activating the SDF-1/CXCR4 signaling pathway,and alleviate myocardial damage in septic rats.
