罗哌卡因调节SIRT1/AMPK通路对妊娠期糖尿病大鼠的保护作用

目的 探讨罗哌卡因调节沉默信息调节因子2相关酶1(SIRT1)/腺苷酸活化蛋白激酶(AMPK)信号通路对妊娠期糖尿病(GDM)大鼠的保护作用.方法 雌性大鼠经高脂高糖喂养8周后与雄性大鼠合笼获得孕鼠,妊娠第6 d腹腔注射STZ制备GDM大鼠模型.将造模成功的大鼠随机分为模型组和罗哌卡因低剂量组(1 mg/kg)、高剂量组(2.0 mg/kg),罗哌卡因高剂量(2.0 mg/kg)+EX-527(1 mg/kg)组,另设对照组,每组15只.全自动生化分析仪检测谷丙转氨酶(GPT)、谷草转氨酶(GOT)和碱性磷酸酶(ALP)活性,以及大鼠血清TC、TG、LDL-C、HDL-C含量;HE染色观察肝组织病理变化;ELISA法检测血清TNF-α、IL-6和IL-1β水平;商品化试剂盒检测肝脏组织MDA、SOD、GSH水平;Western blot检测肝组织Bcl-2、Bax、SIRTl、AMPK蛋白水平.结果 对照组肝组织细胞形态结构正常;与对照组相比,模型组大鼠肝组织有明显脂肪空泡和炎症细胞浸润,血清TG、TC、LDL-C水平、GPT、GOT、ALP水平、TNF-α、IL-6和IL-1β水平、肝组织MDA水平、Bax水平显著升高,血清HDL-C水平、肝组织SOD、GSH水平、Bcl-2、SIRT1、p-AMPK/AMPK蛋白水平显著降低(P<0.05);与模型组相比,罗哌卡因低、高剂量组脂肪空泡显著减少,有少量炎症细胞浸润,血清TG、TC、LDL-C水平、GPT、GOT、ALP水平、TNF-α、IL-6和IL-1 β水平、肝组织MDA水平、Bax水平显著降低,血清HDL-C水平、肝组织SOD、GSH水平、Bcl-2、SIRT1、p-AMPK/AMPK蛋白水平显著升高(P<0.05);与罗哌卡因高剂量组相比,EX-527干预可减弱罗哌卡因对GDM大鼠的保护作用.结论 罗哌卡因通过激活SIRT1/AMPK信号通路可以降低GDM大鼠炎症反应和氧化应激,进而改善GDM症状.
Protective effect of ropivacaine on gestational diabetes mellitus rats by regulating SIRT1/AMPK signaling pathway
Objective To investigate the protective effect of ropivacaine on gestational diabetes mellitus(GDM)rats by regulating the silent information regulator factor 2 related enzyme 1(SIRT1)/AMP-activated protein kinase(AMPK).Methods Female rats were fed with high fat and high sugar for 8 weeks before being caged with male rats to obtain pregnant rats.The sixth day of pregnancy,STZ was injected intraperitoneally to prepare a GDM rat model.The successfully modeled rats were randomly grouped into a model group,a low-dose ropivacaine group(1 mg/kg),a high-dose ropivacaine group(2.0 mg/kg),and a high-dose ropivacaine(2.0 mg/kg)+EX-527(1 mg/kg)group.A control group was also established,with 15 rats in each group.The activities of glutamic pyruvic transami-nase(GPT),glutamic oxaloacetic transaminase(GOT)and alkaline phosphatase(ALP),and the contents of TC,TG,LDL-C and HDL-C in rat serum were detected by automatic biochemical analyzer;HE staining was applied to observe the histopathology of the liver;ELISA method was applied to detect serum levels of TNF-α,IL-6,and IL-1β;the commercial reagent kits were applied to detect the levels of MDA,SOD,and GSH in liver tissue;West-ern blot was applied to detect the levels of Bcl-2,Bax,SIRT1,and AMPK proteins in liver tissue.Results The morphology and structure of liver histiocyte in the control group were normal;compared with the control group,the liver tissue of rats in the model group showed obvious fat vacuoles and infiltration of inflammatory cells,the levels of serum TG,TC,LDL-C,GPT,GOT,ALP,TNF-α,IL-6,and IL-1β,liver tissue MDA,and Bax in rats were ob-viously increased,the levels of serum HDL-C,liver tissue SOD,GSH,Bcl-2,SIRT1,p-AMPK/AMPK were obvi-ously reduced(P<0.05);compared with the model group,the fat vacuoles in the low-dose and high-dose ropiva-caine groups were obviously reduced,with a small amount of inflammatory cell infiltration,the levels of serum TG,TC,LDL-C,GPT,GOT,ALP,TNF-α,IL-6,and IL-1β,liver tissue MDA,and Bax in rats were obviously re-duced,the levels of serum HDL-C,liver tissue SOD,GSH,Bcl-2,SIRT1,p-AMPK/AMPK were obviously in-creased(P<0.05);compared with the high-dose ropivacaine group,EX-527 intervention can reduce the protective effect of ropivacaine on GDM rats.Conclusion Ropivacaine can reduce inflammation and oxidative stress in GDM rats by activating the SIRT1/AMPK signaling pathway,thereby improving GDM symptoms.
