Research on the mechanism of IL-7 on T lymphocyte function in sepsis based on the CD80/CTLA-4 pathway
Objective To explore the influence of IL-7 on T lymphocyte function in sepsis based on the CD80/CTLA-4 pathway.Methods Selection:104 septic patients admitted to our hospital from January 1,2021,to June 6,2023 were chosen.Peripheral blood T lymphocytes were isolated and purified from the patients.The T lym-phocytes were stimulated and cultured,divided into three groups:Control group,IL-7 group,and CYAL-4 group.Flow cytometry was used to detect the apoptosis rate and proliferation index of T lymphocytes.ELISA was used to measure the levels of IL-7,IFN-γ,and TNF-α in lymphocytes.Flow cytometry was used to analyze the expression of CD80 on lymphocyte surface.Immunofluorescence was used to detect the expression of CTLA-4 on T lymphocyte sur-face.Results Compared with the control group,the apoptosis rate of T lymphocytes was significantly reduced in the IL-7 group and CYAL-4 group(45.17±4.02,48.06±4.08,respectively),the proliferation index of T lympho-cytes(6.01±1.04,5.92±1.02,respectively),and the levels of IL-7(115.46±9.07,112.39±9.01,respectively),IFN-y(21.46±2.15,20.07±2.12,respectively),and TNF-α(347.15±2.26,340.87±12.08,respectively)in T lym-phocytes were significantly increased(P<0.05).There was no significant difference in the apoptosis rate,prolifera-tion index,IL-7,IFN-γ,TNF-α levels between the IL-7 group and the CYAL-4 group(P>0.05).Compared with the Control group,the expression of CD80(6.28±0.87)and CTLA-4 positivity(3.68±0.57)in T lymphocytes were significantly decreased in the IL-7 group and CYAL-4 group(P<0.05).There was no significant difference in the ex-pression of CD80 and CTLA-4 positivity between the IL-7 group and the CYAL-4 group(P>0.05).Conclusion IL-7 can inhibit T lymphocyte apoptosis and promote T lymphocyte proliferation in septic patients.It also increases IL-7,IFN-γ,and TNF-α levels in T lymphocytes.The mechanism behind these effects may be associated with the inhi-bition of the CD80/CTLA-4 pathway.
