Effects of E3 ubiquitin ligase TRIM59 on proliferation and invasion of non-small cell lung cancer cells
Objective To explore the effect and mechanism of TRIM59 on proliferation,migration and inva-sion of non-small cell lung cancer cells.Methods Non-small cell lung cancer cell line A549 was divided into siCon-trol group,siTRIM59 group,Flag-Vector group and Flag-TRIM59 group.The cell proliferation activity was detected by CCK-8 assay.The apoptosis rate was detected by TUNEL staining assay.The ability of cell migration and invasion was detected by scratch test and Transwell test.AKT expression and phosphorylation levels were detected by immuno-imprinting assay.Results Compared with siControl group,cell proliferation activity in siTRIM59 group was de-creased(P<0.05),apoptosis ratio was increased(P<0.05),cell mobility was decreased(P<0.05),the number of cells invading the lower chamber of Transwell was decreased(P<0.05),and AKT phosphorylation level was de-creased.Compared with Flag-Vector group,cell proliferation activity was enhanced in Flag-TRIM59 group(P<0.05),apoptosis ratio was decreased(P<0.05),cell mobility was increased(P<0.05),the number of cells invad-ing the lower compartment of Transwell was increased(P<0.05),and AKT phosphorylation level was increased.Conclusion TRIM59 promotes proliferation,migration,and invasion of non-small cell lung cancer cells,as well as AKT phosphorylation.
