目的 探讨癌易感性候选基因2(CASC2)对甲状腺癌细胞(TPC-1)增殖及侵袭转移的影响.方法 收集2020年1月-2022年12月于我院行手术治疗的83例甲状腺癌患者,取甲状腺癌组织及癌旁组织进行石蜡标本制作,并采用实时荧光定量聚合酶链反应(RT-PCR)对其癌组织和癌旁组织中CASC2 mRNA和miR-155-5p RNA的表达水平进行检测.将TPC-1细胞随机分为质粒组、空载组及空白组,质粒组使用脂质体转染试剂转染CASC2a;而空载组转染空载质粒,空白组不做处理.采用CCK-8法检测细胞增殖情况,Transwell侵袭实验检测细胞迁移和侵袭情况.结果 癌组织中lncRNA-CASC2的表达(1.23±0.08)明显低于癌旁组织(2.16±0.11);但miR-155-5p的表达(2.18±0.16)却明显高于癌旁组织(1.46±0.10)(P<0.05),3组细胞 48 h 的A450(分别为 0.32±0.03、0.40±0.04、0.42±0.06)、72 h(分别为 0.65±0.08、0.73±0.10、0.77±0.11)及96 h(分别为0.97±0.12、1.24±0.14、1.35±0.15)间的差异均有统计学意义(P<0.05),且随着时间的增加,3组细胞的A450值均有所增大.比较3组细胞迁移和侵袭能力发现,质粒组细胞的迁移数(54.63±8.95)和侵袭数(33.58±6.63)均明显低于空载组(分别为89.37±10.04、72.37±9.72)和空白组细胞(分别为93.21±10.22、75.82± 9.62)(P<0.05);而空载组和空白组细胞的迁移数和侵袭数差异无统计学意义(P>0.05).结论 LncRNA-CASC2在甲状腺癌组织中呈现低表达,而miR-150-5p在癌组织中呈高表达,两种因子均为参与甲状腺癌的发生发展的关键;且lncRNA-CASC2在细胞水平上可抑制TPC-1细胞的增殖及迁移能力,但其抑癌作用的关键点及调控机制等需进一步探索.
The effect of lncRNA CASC2 on proliferation,invasion and metastasis of thyroid cancer
Objective To explore the effect of cancer susceptibility candidate gene 2(CASC2)on the pro-liferation,invasion,and metastasis of thyroid cancer cell line(TPC-1)cells.Methods Collect 83 thyroid cancer patients who underwent surgical treatment in our hospital from January 2020 to December 2022,and prepare paraf-fin specimens of thyroid cancer and adjacent tissues.Real time fluorescence quantitative polymerase chain reaction(RT PCR)was used to detect the expression levels of CASC2 mRNA and miR-155-5p RNA in the cancer and adja-cent tissues.TPC-1 cells were randomly divided into plasmid group,empty group,and blank group.The plasmid group was transfected with CASC2a using liposome transfection reagent;The empty group was transfected with empty plasmids,while the blank group was not treated.The CCK-8 method was used to detect cell proliferation,while the Transwell invasion experiment was used to detect cell migration and invasion.Results The expression of lncrna-casc2 in cancer tissues(1.23±0.08)was significantly lower than that in adjacent tissues(2.16±0.11);However,the expression of mir-155-5p(2.18±0.16)was significantly higher than that of the adjacent tissues(1.46±0.10)(P<0.05).There were a significant difference in A450 values at 48 h(0.32±0.03,0.40±0.04,0.42±0.06,respectively),72 h(0.65±0.08,0.73±0.10,0.77±0.11,respectively)and 96 h(0.97±0.12,1.24±0.14,1.35±0.15,respectively)in the cell group(P<0.05);And with the increase of time,the A450 value of the three groups of cells increased.The migration and invasion abilities of the three groups were compared.The migration number(54.63±8.95)and invasion number(33.58±6.63)of the plasmid group were significantly lower than those of the empty group(89.37±10.04,72.37±9.72,respectively)and the blank group(93.21±10.22,75.82±9.62,respectively)(P<0.05);However,there was no significant difference in the number of migration and invasion between the empty group and the blank group(P>0.05).Conclusion LncRNA-CASC2 exhibits low expression in thyroid cancer tissue,while miR-150-5p is highly expressed in cancer tissue.Both factors are crucial for the occurrence and development of thyroid cancer;Moreover,lncRNA-CASC2 can inhibit the proliferation and migration ability of TPC-1 cells at the cellular level,but the key points and regulatory mechanisms of its anti-tumor effect need further exploration.
