Study on the biological function of RANBP17 in glioblastoma
Objective To investigate the biological function ofRAN-binding protein 17(RANBP17)gene in glioblastoma(GBM).Methods The expression and prognostic value of RANBP17 in GBM was analyzed using UALCAN and CGGA databases.The LinkedOmics database was used to screen the genes related to RANBP17 ex-pression,and the gene function enrichment analysis was performed through the DAVID database.CCK8,flow cytom-etry,and Transwell assays were conducted to evaluate the impact of RANBP17 on the proliferation,apoptosis,mi-gration and invasion in GBM U251 cell.ENCORI,miRWalk and miRDB databases were utilized to predict upstream miRNA of RANBP17.Results The expression level of RANBP17 in GBM was lower than that in normal tissues(P<0.05).The expression level of RANBP17 was highly expressed in GBM patients under 41 years of age,secondary GBM,IDH mutant,MGMT promoter methylation and 1p19q co-deletion(all P<0.01).The enrichment analysis of 500 RANBP17 expression related genes showed that RANBP17 expression related genes can participate in inflamma-tory response,cell adhesion,negative regulation of apoptosis,tight junction and linoleic acid metabolism and other processes and signaling pathways.Cell functional experiments in vitro demonstrated that overexpression of RANBP17 decreased cell proliferation,migration,invasion and facilitated apoptosis in GBM U251 cell(P<0.05).There were 8 common miRNAs in ENCORI,miRWalk and miRDB databases(hsa-miR-650,hsa-miR-361-5p,hsa-miR-421,hsa-miR-9-5p,hsa-miR-3612,hsa-miR-299-5p,hsa-miR-2115-3p and hsa-miR-19b-3p)regulate RANBP17.Con-clusions RANBP17 may be involved in the occurrence and development of GBM as a tumor suppressor gene,which is a potential prognostic biomarker and therapeutic target in GBM.
