Effects of tonsillectomy on TH1/TH2 immune balance,B cell subsets and LILR expression in rats
Objective To analyze the effects of tonsillectomy on TH1/TH2 immune balance and immune protein expression in rats and its possible mechanism.Methods A total of 21 adult male rats were selected and di-vided into control group,intracapsular group and extracapsular group using random number table method.The rats in the control group were fed normally,the rats in the intracapsular group were given intracapsular resection,and the rats in the extracapsular group were given extracapsular resection.The levels of TH1/TH2 cytokines,TH1/TH2,CD3+,CD4+,CD8+cells and immune protein in each group were compared,and the expression of MCP-1 and CCR2 proteins in each group were detected.Results The levels of TH 1,TH2 and TH 1/TH2 in intracapsular group and ex-tracapsular group were lower than those in control group,and those in intracapsular group were higher than those in extracapsular group.The levels of CD3+,CD4+,CD8+and CD4+/CD8+in intracapsular group and extracapsular group were lower than those in control group,and those in intracapsular group were higher than those in extracapsular group.The levels of IgA,IgG and IgM in intracapsular and extracapsular groups were lower than those in control group,and the levels in intracapsular and extracapsular groups were higher than those in control group.Compared with the control group,the change of B cell subsets in the intracapsular group was lower than that in the extracapsu-lar group.Intracapsular resection had less effect on B-cell subsets and B1 was expressed on each B-cell subset,while the expression of B2 and B3 on B-cell subsets was decreased in extracapsular group.The expression of B4 was unique on ASC,and the level of B4 in the extracapsular group was lower than that in the intracapsular group.Com-pared with the average fluorescence intensity,the level of B1 and B4 in the intracapsular group was significantly higher than that in the extracapsular group.The relative expression levels of MCP-1 and CCR2 protein in intracapsu-lar group and extracapsular group were higher than those in control group,and those in intracapsular group were low-er than those in extracapsular group,with statistical significance(P<0.05).Conclusion Both intracapsular and ex-tracapsular tonsillectomy rats showed immune imbalance and decreased immune cells and immune proteins,but in-tracapsular tonsillectomy could improve TH1/TH2 cytokines,TH1/TH2immune balance,immune protein expres-sion,CD3+,CD4+,CD8+and CD4+/CD8+levels in rats.The mechanism may be related to the regulation of MCP-1/CCR2 protein pathway.
