miR-551在甲状腺癌患者中的表达及靶向ERBB4对SW579细胞移植瘤生长的影响

目的 探讨miR-551在甲状腺癌患者中的表达及靶向ERBB4对SW579细胞移植瘤生长的影响.方法 选取2019年1月-2022年3月于我院病理科采集并保存的甲状腺乳头状癌组织及其配对的癌旁组织 100 例.将 SW579 细胞随机分为 Control 组、miR-NC 组、miR-551 组和 miR-551+pcDNA-ERBB4 组.RT-PCR检测甲状腺癌组织和细胞中miR-551表达,分析甲状腺癌组织中miR-551表达和临床病理特征相关性;CCK-8、Transwell法、流式细胞仪分别检测细胞增殖、侵袭和凋亡率;蛋白质印迹法检测细胞中ERBB4蛋白表达;荧光素酶活性基因报告检测miR-551和ERBB4靶向关系.建立裸鼠皮下移植瘤模型,分为miR-NC组和miR-551组,比较两组裸鼠肿瘤体积;TUNEL检测肿瘤组织细胞凋亡率.结果 甲状腺癌组织中miR-551表达低于癌旁组织(P<0.05).与甲状腺上皮细胞相比(1.00±0.10),甲状腺癌细胞中miR-551表达(0.32±0.04)明显降低(P<0.05).与Control组相比,miR-551组细胞中miR-551表达(1.28±0.13)和细胞凋亡率(25.72±1.78)明显增加,24h、48h细胞活力(分别为52.38±5.46、32.29±3.17)、细胞侵袭数目(81.06±6.32)和细胞中ERBB4蛋白(0.46±0.05)表达明显降低(P<0.05);与miR-551组相比,miR-551+pcDNA-ERBB4组细胞中miR-551表达和细胞凋亡率(7.69±0.82)明显降低,24h、48h细胞活力(分别为85.11±7.61、71.56±6.82)、细胞侵袭数目(136.82±7.54)和细胞中 ERBB4 蛋白表达(0.84±0.08)明显增加(P<0.05);与 miR-NC 组(1.00±0.10)相比,转染野生型ERBB4(ERBB4-WT)时miR-551组荧光素酶活性明显降低(0.34±0.04)(P<0.05);与miR-NC组相比,miR-551组裸鼠肿瘤体积明显减少,肿瘤组织中细胞凋亡率(51.62±4.31)明显升高(P<0.05).结论 甲状腺癌组织中miR-551呈低表达,过表达miR-551可靶向调控ERBB4抑制甲状腺癌细胞增殖和侵袭,诱导甲状腺癌细胞凋亡.
The expression of miR-551 in thyroid cancer patients and the mechanism of targeting ERBB4 on the growth of SW579 cell transplanted tumor
Objective To explore the expression of miR-551 in thyroid cancer patients and the effect of tar-geted ERBB4 on the growth of SW579 cell transplantation tumors.Method 100 cases of papillary thyroid cancer tissue and its paired adjacent tissues collected and preserved in the Department of Pathology of our hospital from Jan-uary 2019 to March 2022 were selected.SW579 cells were randomly divided into Control group,miR-NC group,miR-551 group,and miR-551+pcDNA-ERBB4 group.RT-PCR was used to detect the expression of miR-551 in thyroid cancer tissue and cells,and to analyze the correlation between miR-551 expression and clinical pathological features in thyroid cancer tissue;CCK-8,Transwell method,and flow cytometry were used to detect cell proliferation,inva-sion,and apoptosis rates,respectively;Protein blotting was used to detect the expression of ERBB4 protein in cells;The luciferase activity gene report detects the targeting relationship between miR-551 and ERBB4.Establish a nude mouse subcutaneous transplant tumor model and divide it into miR-NC group and miR-551 group.Compare the tumor volume between two groups of nude mice;TUNEL was used to detect the apoptosis rate of tumor tissue cells.Result The expression of miR-551 in thyroid cancer tissue was lower than that in adjacent tissues(P<0.05).Com-pared with thyroid epithelial cells(1.00±0.10),the expression of miR-551 in thyroid cancer cells was significantly reduced(0.32±0.04)(P<0.05).Compared with the control group,the expression of miR-551(1.28±0.13)and apoptosis rate(25.72±1.78)in the miR-551 group cells were significantly increased,while 24 h and 48 h cell viabil-ity(52.38±5.46,32.29±3.17,respectively),cell invasion number(81.06±6.32),and ERBB4 protein(0.46±0.05)in the cells were significantly reduced(P<0.05);Compared with the miR-551 group,the miR-551+pcDNA-ERBB4 group showed a significant decrease in miR-551 expression and apoptosis rate(7.69±0.82),while 24 h and 48 h cell viability(85.11±7.61,71.56±6.82,respectively),cell invasion number(136.82±7.54),and expression of ERBB4 protein in cells(0.84±0.08)were significantly increased(P<0.05).Compared with the miR-NC group(1.00±0.10),the luciferase activity of the miR-551 group significantly decreased(0.34±0.04)when transfected with wild-type ERBB4(ERBB4-WT)(P<0.05).Compared with the miR-NC group,the miR-551 group showed a signifi-cant decrease in tumor volume and an increase in cell apoptosis rate in tumor tissue(51.62±4.31)(P<0.05).Con-clusion miR-551 is downregulated in thyroid cancer tissues.Overexpression of miR-551 can target and regulate ERBB4,inhibiting proliferation and invasion of thyroid cancer cells while inducing apoptosis.
