高频重复经颅磁刺激对血管性痴呆大鼠学习记忆能力的影响及机制

目的 观察血管性痴呆(VD)大鼠学习记忆能力是否通过高频重复经颅磁刺激(HrTMS)发生改变,并探讨其作用机制.方法 Wistar大鼠83只,5只用于测定TMS静息运动阈值,另外78只大鼠随机分成假手术组、模型组和HrTMS组.用改良四血管闭塞法(4VO)制作全脑缺血再灌注模型;术后第3天开始给予HrTMS.大鼠空间学习记忆能力检测采用Morris水迷宫实验;观察海马形态学改变使用苏木精-伊红(HE)染色,免疫组化技术检测BDNF、MAP2蛋白.结果 与模型组逃逸潜伏期(56.67±0.72)s相比,HrT-MS组(52.51±0.70)s显著缩短(P<0.05);在空间探索实验中,大鼠在60 s内穿过原有平台的次数,HrTMS组(2.13±0.30)次多于模型组(1.00±0.27)次(P<0.05).HE染色显示,相比模型组,HrTMS组CA1区细胞坏死数量减少,形态学改变减轻.免疫组化提示,rTMS治疗14 d后,HrTMS组BDNF COD值(0.680±0.019)、MAP2 COD 值(0.456±0.020)与模型组的BDNF COD 值(0.606±0.019)、MAP2 COD 值(0.345±0.020)相比,表达增高(P<0.05).结论 高频rTMS能上调BDNF、MAP2蛋白的表达,可能是其促进血管性痴呆大鼠学习记忆认知功能恢复的原因.
Effects of High-frequency repetitive transcranial magnetic stimulation on learning and memory in vas-cular dementia rats and its mechanism
Objective To observe the effect of high-frequency transcranial magnetic stimulation(HrTMS)on learning and memory abilities in vascular dementia(VD)rats and explore its mechanism.Methods 5 of 83 male Wistar rats were used for magnetic stimulation resting motion threshold measurement.The other 78 rats were divided into SHAM group,MODEL group,and HrTMS group by randomly.The whole brain ischemia-reperfusion model was made by modified four-vessel occlusion(4VO);HrTMS was administered on the 3rd day after surgery.Morris water maze test was used to detect the spatial learning and memory ability of rats;HE staining technique was used to ob-serve the morphological changes of hippocampus,BDNF and MAP2 proteins were detected by immunohistochemis-try.Results Compared with the escape latency of the model group(56.67 s±0.72 s),the HrTMS group(52.51 s±0.70 s)was significantly shortened(P<0.05);In the space exploration experiment,the number of times rats crossed the original platform within 60 seconds was higher in the HrTMS group(2.13±0.30)than in the model group(1.00±0.27)(P<0.05).HE staining showed that compared to the model group,the HrTMS group had a reduced number of cell necrosis and reduced morphological changes in the CA1 region.Immunohistochemical analysis showed that after 14 days of rTMS treatment,the expression of BDNF COD(0.680±0.019)and MAP2 COD(0.456±0.020)in the HrTMS group increased compared to the BDNF COD(0.606±0.019)and MAP2 COD(0.345±0.020)values in the model group(P<0.05).Conclusion High frequency rTMS can upregulate the expression of BDNF and MAP2 pro-teins,which may be the reason why it promotes the recovery of learning,memory,and cognitive function in VD rats.
