Expression and role of lncRNA LINC01133 in invasive periodontitis
Objective:To explore the role and mechanism of lncRNA LINC01133 in invasive periodontitis.Methods:The expressions of LINC01133 and matrix metalloproteinase-9(MMP-9)in gingival tissues of different groups of healthy population and invasive periodontitis patients under different inflammatory conditions were detected by qRT-PCR,and IL-6 and TNF-α inflammatory cytokines were detected by ELISA.The invasive periodontitis cell model was established and human gingival fibroblasts(HGFs)were stimulated by lipopolysaccharide of Escherichia coli.The expression of LINC01133 in HGFs cells was detected in vitro.The LINC01133 overexpression vector was constructed to transfect HGFs,and the changes of mRNA and protein secretion of the above factors in the HGFs were detected by qRT-PCR and Western blotting.Results:Low expression of LINC01133 and high expression of MMP-9,IL-6,and TNF-α were found in human gingival tissues.There was an inverse correlation between LINC01133 and MMP-9.After the construction of inflammatory cell model,it was found that LINC01133 showed a low expression.Furthermore,after transfection the expression of LINC01133 was upregulated in the HGFs,and it was found to inhibit the expression of MMP-9,IL-6 and TNF-α,thus reducing the occurrence of inflammation and improving the progression of invasive periodontitis.Conclusions:The low expression of LINC0113 in inflammatory gingival tissues suggests its involvement in the pathogenesis of invasive periodontitis.Moreover,LINC01133 can regulate the expression of MMP-9,IL-6,and TNF-α to control the progression of invasive periodontitis.
