目的:探讨通窍汤加减对过敏性鼻炎(AR)模型大鼠血清可溶性程序性死亡配体1(sPD-L1)、T细胞免疫球蛋白域黏蛋白域蛋白-1(Tim-1)表达的影响.方法:SPF级SD大鼠,建模成功后分为模型组、低剂量组、高剂量组以及地塞米松组.检测各组大鼠行为学评分,ELISA法检测IFN-γ、IL-4水平,H-E染色观察大鼠鼻黏膜组织病理学形态结构,Real time-PCR及免疫印迹检测sPD-L1、Tim-1 mRNA及蛋白表达.结果:与健康组相比,模型组行为学评分、IL-4水平、Tim-1 mRNA及蛋白表达均升高,IFN-γ水平、sPD-L1 mRNA及蛋白表达均降低;与模型组相比,低、高剂量组及地塞米松组行为学评分、IL-4水平、Tim-1 mRNA及蛋白表达均降低,IFN-γ水平、sPD-L1 mRNA及蛋白表达均升高;与低剂量组相比,高剂量组与地塞米松组行为学评分、IL-4水平、Tim-1 mRNA及蛋白表达均降低,IFN-γ水平、sPD-L1 mRNA及蛋白表达均升高;与高剂量组相比,地塞米松组行为学评分、IL-4水平、Tim-1 mRNA及蛋白表达均升高,IFN-γ水平、sPD-L1 mRNA及蛋白表达均降低.模型组可见鼻黏膜下层水肿,并有大量炎症细胞浸润;与模型组相比,低剂量组、高剂量组以及地塞米松组均有所改善.结论:通窍汤加减对模型AR大鼠具有提高血清sPD-L1水平,以及降低Tim-1水平和调控Th1/Th2水平比例的功效.
Effect of modified Tongqiao decoction on expression of sPD-L1 and Tim-1 in serum of allergic rhinitis model rats
Objective:To study the effects of Tongqiao decoction on the expression of soluble programmed death ligand-1(sPD-L1)and T cell immunoglobulin domain and mucin domain protein 1(Tim-1)in serum of allergic rhinitis(AR)model rats.Methods:After successful modeling,SPF grade SD rats were divided into model group,low-dose Tongqiao decoction group,high-dose Tongqiao decoction group,and dexamethasone group.Behavioral scores of rats in each group were assessed.The levels of IFN-γ and IL-4 were measured using the ELISA method.The pathological morphological structure of the rat nasal mucosa tissue was observed using H-E staining.The expression of sPD-L1 and Tim-1 mRNA and proteins was detected by Real-time PCR and Western blotting.Results:Compared with the healthy group,the model group showed an increase in behavioral scores,IL-4 levels,Tim-1 mRNA and protein expression,and IFN-γ levels,sPD-L1 mRNA and protein expression were all reduced.Compared with the model group,the behavioral scores,IL-4 levels,Tim-1 mRNA and protein expression were all reduced,and the IFN-γ level,sPD-L1 mRNA and protein expression all increased in the low dose and high dose Tongqiao decoction groups,as well as the dexamethasone group;Compared with the low-dose group,the high-dose Tongqiao decoction group and dexamethasone group showed decreased behavioral scores,IL-4 levels,Tim-1 mRNA and protein expression,and increased IFN-γ levels,sPD-L1 mRNA and protein expression.Compared with the high-dose Tongqiao decoction group,the dexamethasone group showed an increase in behavioral scores,IL-4 levels,Tim-1 mRNA and protein expression,and a decrease in IFN-γ levels,sPD-L1 mRNA and protein expression.The model group showed edema in the submucosal layer of the nose,with a large amount of inflammatory cell infiltration.Compared with the model group,the low-dose Tongqiao decoction group,high-dose Tongqiao decoction group,and dexamethasone group all showed improvement.Conclusion:The modified Tongqiao decoction was found to improve the serum sPD-L1 level,reduce the Tim-1 level,and regulate the Th1/Th2 ratio in AR rats.
