Honeysuckle vine extract activates the Wnt/β-catenin pathway to promote osteoporotic fracture healing
Objective:To investigate the effects of honeysuckle vine extract on rat models of osteoporotic fractures and its mechanism.Methods:The rats were randomly divided into Control group,Model group,and low-dose(Low),medium-dose(Medium),and high-dose(High)honeysuckle vine extract groups.Modeling was performed in all groups except for the Control group.After successful modeling,the Low,Medium,and High groups were given 50,100,and 200 mg·kg-1·d-1 honeysuckle vine extract by gavage for 12 weeks,while the Control group and Model group were given equal volume of normal saline.The dual energy X-ray radiography results,callus H-E staining results,bone density,serum alkaline phosphatase(ALP)levels,and serum inflammatory cytokine[tumor necrosis factor-alpha(TNF-α),interleukin-1 beta(IL-1β)]levels were measured and compared between the groups.Western blotting and reverse transcription-polymerase chain reaction(RT-PCR)were used to detect the Wnt3,β-catenin protein,and mRNA expression levels in callus tissues.Results:The results of dual energy X-ray radiography showed that the fracture line of the Model group was clear,and there was evident callus growth.Compared with the Model group,the Low,Medium and High groups showed reduced gap at the fracture site and smaller callus volume.Among them,the High group exhibited the best fracture healing.H-E staining results showed that chondrogenic callus was observed in the Model group,with fewer mature trabecular cells and disordered bone trabeculae.In the Low,Medium and High groups,the histopathological changes were improved to varying degrees,with an increasing number of trabeculae and their arrangement became more orderly.Compared with the Control group,the Model group of rats showed significant decreases in bone density,ALP levels,as well as expression levels of Wnt3 and β-catenin proteins and mRNA.Additionally,serum levels of TNF-α and IL-1β were significantly elevated.Compared with the Model group,the Low,Medium,and High groups showed significant increases in bone density,ALP levels,as well as expression levels of Wnt3 and β-catenin proteins and mRNA.Additionally,serum levels of TNF-α and IL-1β were significantly decreased.Conclusion:The extract of honeysuckle vine can promote fracture healing in osteoporotic fracture rat models,and its mechanism may be related to its ability to activate the Wnt/β-catenin pathway.
