Impact of lncRNA EBLN3P on the malignant biological behavior of non small cell lung cancer cells by regulating miR-340-5p/mitochondrial transcription factor A axis
Objective:To investigate the impact and mechanism of long non-coding RNA EBLN3P on the malignant biological behavior of non small cell lung cancer(NSCLC)cells by regulating microRNA-340-5p(miR-340-5p)/mitochondrial transcription factor A(TFAM)axis.Methods:NSCLC cell lines were applied as research objects.A549 cells were grouped into ctrl group,si-NC group,si-EBLN3P group,si-EBLN3P+NC inhibitor group,and si-EBLN3P+miR-340-5p inhibitor group.Cell proliferation,migration,and invasion were detected by MTT,EdU,Transwell respectively.The targeting relationship among EBLN3P,miR-340-5p,and TFAM was verified by dual luciferase reporter assay,and expression of TFAM,miR-340-5p,E-cadherin,and N-cadherin proteins was detected by qRT-PCR and Western blotting respectively.Results:The expression of EBLN3P was up-regulated in lung cancer tissues and cells.In NSCLC cells,the expression of TFAM was up-regulated and the expression of miR-340-5p was decreased.Inhibiting the expression of EBLN3P could obviously inhibit the proliferation,migration,and invasion of NSCLC cells,and the expression of E-cadherin protein increased,while the expression of N-cadherin protein decreased.There was a targeting relationship between EBLN3P,TFAM and miR-340-5p.Conclusion:EBLN3P is highly expressed in NSCLC tissues and cells,inhibiting the expression of EBLN3P can inhibit the proliferation,migration,and invasion of non small cell lung cancer cells by regulating the miR-340-5p/TFAM signal axis.
