Objective:To investigate the effect and mechanism of miR-126-5p on cisplatin resistance,proliferation and invasion of thyroid cancer cells.Methods:The expression of miR-126-5p and MAPK1 mRNA in the cells were detected by RT-qPCR.The cells were divided into control group,miR-126-5p mimic group,miR-NC group,miR-126-5p mimic+pc-MAPK1group,miR-126-5p mimic+pc-NC group.The cell cisplatin(CDDP)resistance and proliferation were measured by CCK-8 assay,and invasion was measured by Transwell,the targeting relationship was verified by the dual-luciferase reporter assay,and expression level of MAPK1 protein was detected by Western blotting.Results:Compared with normal tissues and cells,the mRNA level of miR-126-5p was significantly down-regulated in thyroid cancer tissues and cells.Compared with the thyroid cancer cell line TPC-1,the expression of miR-126-5p in thyroid cancer cisplatin-resistant cell line TPC-1/CDDP was significantly down-regulated.Either in TPC-1 cells or TPC-1/CDDP cells,compared with the control group,the IC50 value of CDDP,cell proliferation,and the number of invasive cells per visual field of the miR-126-5p mimic group cells were all significantly reduced.The miR-126-5p targeted the down-regulation of MAPK1.Overexpression of MAPK1 reversed the effect of miR-126-5p overexpression on the IC50 value of cell CDDP,cell proliferation and the number of invasive cells per field.Conclusion:The miR-126-5p overexpression enhances the cisplatin sensitivity of TPC-1 and TPC-1/CDDP cells by targeting the down-regulation of MAPK1,and inhibits cell proliferation and invasion.
