Objective:To explore the expression of small nucleolar RNA host genes 15(SNHG15)and LINC00261 in nasopharyngeal carcinoma tissues and their clinical significance.Methods:Tumor tissue and adjacent tissue samples of 93 cases of nasopharyngeal carcinoma undergoing radical resection in the Department of Otolaryngology of our hospital from January 2018 to February 2020 were collected,as the well as clinical data of patients;The cultured nasopharyngeal carcinoma CNE-1 cells and normal nasopharyngeal epithelial NP69 cells were divided into blank control group,shRNA-NC group(transfected with SNHG15 control),SNHG15 shRNA group(transfected with SNHG15),pcDNA-NC group(transfected with LINC00261 control),and LINC00261 pcDNA group(transfected with LINC00261);RT-qPCR was used to detect the expression of SNHG15 and LINC00261 in CNE-1 cells.CKK-8 method and clone formation assay were used to detect the proliferation of cells in each group.Pearson method was applied to analyze the correlation between the expression of SNHG15 and LINC00261 in nasopharyngeal carcinoma tissues.Kaplan-Meier method was applied to analyze the relationship between the expression of SNHG15 and LINC00261 in nasopharyngeal carcinoma tissues and prognosis.Multivariate Cox regression analysis was applied to investigate the factors affecting the prognosis of nasopharyngeal carcinoma patients.Results:The expression of SNHG15 in nasopharyngeal carcinoma tissues was higher than that in adjacent tissues,while the expression of LINC00261 was lower than that in adjacent tissues.The expression of SNHG15 and LINC00261 in nasopharyngeal carcinoma tissue were negatively correlated.The expression of SNHG15 in pharyngeal cancer cells was higher than that in normal nasopharyngeal epithelial cells,while the expression of LINC00261 was lower than that in normal nasopharyngeal epithelial cells.The number of cell clones formed in the SNHG15-shRNA group and LINC00261-pcDNA group was lower than that in the control group,shRNA-NC group,and pcDNA-NC group,indicating a decrease in cell proliferation ability,and there was no statistically significant difference in age,tumor diameter,and histological type among nasopharyngeal carcinoma patients with different levels of SNHG15 and LINC00261 expression,while the proportions of patients with lymph node metastasis and TNM stage Ⅲ+Ⅳ in the SNHG15 high expression group and LINC00261 low expression group were higher than those in the SNHG15 low expression group and LINC00261 high expression group.The three-year survival rate(84.78%)of the SNHG15 low expression patients was higher than that of the SNHG15 high expression patients(65.95%),while the three-year survival rate(63.04%)of the LINC00261 low expression patients was lower than that of the LINC00261 high expression patients(87.23%).The lymph node metastasis,TNM staging,and SNHG15 were risk factors for death in nasopharyngeal carcinoma patients,while LINC00261 was a protective factor for such patients.Conclusion:The expression of SNHG15 and LINC00261 in nasopharyngeal carcinoma tissues are closely related to the clinical pathological characteristics and prognosis of patients.
