白纹伊蚊气味受体基因OR21a的克隆及定量分析

本文克隆白蚊伊蚊气味受体基因OR21a并分析其是否与搜寻宿主或吸血行为相关.通过设计简并引物,PCR扩增得到白纹伊蚊气味受体基因OR21a片段,采用SYBR荧光定量PCR方法,以白纹伊蚊β-肌动蛋白基因作为内参,比较了OR21a基因在未吸血雌蚊、雄蚊、打断吸血雌蚊和饱血雌蚊头部表达量的差异.结果显示,白纹伊蚊OR21a基因在未吸血雌蚊头部表达量最高,饱血雌蚊头部表达量最低,而在雄蚊头部的表达量介于打断吸血雌蚊和饱血雌蚊之间.本研究发现白蚊伊蚊会依据不同的生理需求改变OR21a基因的表达量,推测可能与白纹伊蚊搜寻宿主及吸血行为有一定的相关性.
CLONING AND EXPRESSION ANALYSIS OF ODORANT RECEPTOR GENE (OR21A) IN AEDES ALBOPICTUS
Sequence of the odorant receptor gene (OR21a) of Aedes albopictus were identified by using degenerate primers.Expression of OR21a mRNA was investigated by quantitative Real-Time RCR in the heads of adult male,interrupt sucking female,engorged and non-feeding adult female.Levels of OR21 a mRNA were normalized to mRNA encoding β-action.The results showed that OR21 a were differentially expressed in the heads of adult male,feeding-interrupted female,engorged and non-feeding adult female.The expression level of non-feeding adult female,feeding-interrupted female,adult male and engorged adult female was reduced successively.The expression level of OR21a changed for need.Maybe the OR21a gene has relationship with the behavior of blood feeding.
