Effect of morphine on the expression of survivin and livin in human breast cancer cells
Objective; To investigate the effect of morphine on proliferation and apoptosis in human breast cancer cell line MCF-7, with a focus on the expression of survivin and livin . MCF-7 cells were seeded in 6-well plates(1 ×103 /ml) and divided into 3 groups; group I normal control(group C) , group Ⅱ was exposed to 0.1 μmol/L morphine ( group Ml ) , group Ⅲ was exposed to 1. 0 μmol/L morphine ( group M2). The expression of survivin and livin in different group of cells was detected by RT -PCR and Western blot at 24 hours of incubation with morphine. The apoptosis of cells was determined by flow cytometry at 24 hours of incubation with morphine. The proliferation of cells was determined by Thiazolyl blue tetrazolium bromide (MTT) method at 24,48, and 72 hours of incubation with morphine respectively. Results; Compared with group C, the expression of survivin and livin , and the proliferation level of group M1 and M2 was decreased significantly ( P < 0. 01) , however, the apoptosis rate of cells was increased significantly ( P <0. 01). Conclusion; Morphine could inhibit the proliferation of the cells and accelerate the cell apoptosis through down-regulating the expression of survivin and livin genes in human breast cancer cell line MCF -7.
