To accelerate the breeding progress of Cucurbita moschata Duch.(C.moschata),the study aimed to de-velop molecular markers which are tightly linked with quantitative trait locus(QTL)of lutein content in C.moschata and are highly practicable.Eight pairs of insertion-deletion(InDel)molecular markers were developed between the two flanking molecular markers R1 38695 and R2 58819,based on the previous localization of the primary QTL locus qlut11-a which was tightly linked to lutein content in F2 population.Genotype analysis and phenotypic analysis of lutein content for the F2 population and recombinant in-breed lines(RILs)in part of the high generation(F8)re-vealed that,InDel molecular markers G005310 and G005670 could be used to effectively screen materials with high and low lutein contents.Furthermore,the two InDel molecular markers were proved to be able to culture pump-kin germplasms with high lutein content in the construction of near isogenic line(NIL).The highest lutein content in BC5.F1 fruit was about 2.8 times as high as the lutein content in parent fruit with low lutein content,and the lutein content in BC5 F1 fruit accounted for about 96% of the lutein content in parent fruit with high lutein content.The research results can provide more practical molecular markers for accelerating breeding process of pumpkin germplasms with high lutein content.
