Analysis of the expression pattern of structural genes and transcription factors encoding genes related to the anthocyanin synthesis in six cultivars of Prunus mume Cinnabar Purple Group
To clarify the molecular regulatory mechanism for the formation of phenotypic differences in flower color of Prunus mume Cinnabar Purple Group,six varieties of P.mume Cinnabar Purple Group with gradient changes in flower color from light to dark were used as the test materials.The floral color phenotypes and anthocyanin contents of petals of different varieties at full-bloom stage were determined by colorimeter and spectrophotometer,respectively.The tran-scriptional characteristics of the structural genes for antho-cyanin synthesis(PmCHS,PmCHI,PmF3H,PmF3′H,PmDFR,PmANS,PmUFGT)and the transcription factors encoding genes(PmMYB1,PmbHLH3)were analyzed by qRT-PCR and correlation analysis.The results showed that the change trend of anthocyanin content in petals among the varieties was consistent with that of flower color redness.The rela-tive expression of PmMYB1,PmbHLH3,PmF3′H,PmDFR and PmUFGT were highly significantly and positively correlated with anthocyanin content.The relative expression of transcription factors encoding genes PmMYB1 and PmbHLH3 were high-ly significantly and positively correlated,and all of them were significantly or highly significantly and positively correlated with the expression levels of the structural genes PmF3′H,PmDFR and PmUFGT.It is hypothesized that the transcription factors encoding genes PmMYB1 and PmbHLH3 regulate the expression of the structural genes PmF3′H,PmDFR and PmUFGT and thus regulate the color presentation of the petals of P.mume Cinnabar Purple Group.
