Analysis of codon use features of APN gene in pigs
The objective of our study is to understand APN gene codon use features and provide guidance and basis for selecting appropriate expression systems.In this research,pig APN gene was analyzed by EMBOSS software package,and then compared with genomes in model organisms and APN genes from 14 animal species.The results showed that most of the pig APN genes were bias toward the synonymous codons with G and C at the third codon position.There were twenty-three biased codons (RSCU> 1).The comparative analysis of APN genes from different animal species showed that gene expression level was general in 14 animal species.RSCU and CDS clustering analysis showed that the phylogenetic tree based on CDS was closer to the real system classification of 14 species.The differences in codon usage frequency between pig APN gene and genome of Mus musculus were less than those in other two kinds of model organisms' genome.This study could provide certain theoretical basis for selecting the appropriate receptor species in animal genetic improvement,selecting appropriate expression systems and improving the expression level of APN gene.
