花生过敏原蛋白Ara h2的分离纯化研究

食物过敏是食品安全领域内比较突出的问题,花生因其高致敏性被列为八大致敏原之首.在花生致敏原中,Ara h2是主要致敏原.以Ara h2为研究对象,通过对花生脱脂,设置不同浸提液、pH、料液比、浸提时间的蛋白质浸提条件,透析分离纯化目标蛋白质,并用十二烷基磺酸钠-聚丙烯酰胺凝胶电泳对优化结果进行鉴定,正交试验确定最佳浸提工艺.结果表明:最佳浸提工艺参数为浸提时间90 min、浸提pH值7.4、浸提料液比1:8、浸提液Tris-HC1缓冲液.在此优化条件下目标蛋白质含量可达1.58mg· mL-1,浸提率可达47.8%.
Isolation and purification of peanut allergen protein Ara h2
Food allergy is a prominent food safety problems,and peanut allergens were classified into the first of the eight allergens because of its high allergenicity.In peanut allergen,Ara h2 is one of the major allergens.In this experiment,Ara h2 as the research object,based on the defatted peanut,setting different extracts,pH,material liquid ratio,extraction time of the protein extraction conditions,the target protein was purified with dialysis,and twelve sodium dodecyl sulfate polyacrylamide gel electrophoresis was used to identify the optimization results,and the optimum extraction process was determined by the orthogonal test.The results showed that the optimum extraction parameters were:extraction time 90 min,pH 7.4,liquid ratio 1:8,Tris-HCl buffer.Under this method,the content of target protein can reach 1.58 mg · mL-1,and the yield can reach 47.8%.
