Construction of gene mutation library of Acinetobacter baumannii and screening and identification of drug resistance suppressor genes
In order to explore the regulatory genes regulating drug resistance of Acinetobacter baumannii,the mutant library of sensitive A.baumannii strain AB43 was established by Tn5 transposable mutation technique,and the antibiotic screening models of imipenem(IPM)and tetracycline(Tet)were constructed.Finally,the gene mutation sites of drug resistant mutants were identified by whole genome sequencing.The results showed that six mutants with increased resist-ance to imipenem and one mutant with increased resistance to tetracycline were successfully screened from more than 4 000 mutants in the mutation bank.Genome-wide sequencing confirmed that mutations in UvrD,LipA,CzcD,OprD and other genes led to changes in AB43 resistance,biofilm formation ability and growth rate of A.baumannii.The results of this study lay a foundation for further exploring the regulation mechanism of drug resistance of A.baumannii.
