Study on the therapeutic effect of Cartiex? type Ⅱ collagen on osteoarthritis
To investigate the therapeutic effect of Cartiex™ type Ⅱ collagen on osteoarthritis(OA)induced by sodium iodoace-tate in rats.The OA model was established by a single injection of iodoacetate sodium solution into the knee joint of rats.After modeling,the rats were treated with type Ⅱ collagen and bovine bone collagen by gavage.Diet,water intake,body weight and mechanical plantar pain were measured weekly during the experiment.At the end of the experiment,blood samples were col-lected from the heart and the contents of PGE-2,TGF-β,MMP-13,IL-1β,IL-6 and TNF-α in serum were detected.The knee tissues were fixed with formaldehyde and decalcified for pathological examination.The results showed that the knee joints of the model rats showed obvious swelling during the experiment,which was relieved in different degrees in each drug treatment group compared with the OA model group.Compared with the OA model group,the high and low dose groups treated with type Ⅱcollagen could significantly increase the mechanical pain value of rats,and significantly reduce the serum levels of MMP-13,IL-1β,IL-6,TNF-α,but significantly increase the contents of PGE-2 and TGF-β in serum.After the intervention of type Ⅱ collagen,the arthritis symptoms of rats were significantly improved,cartilage tissue hyperplasia,inflammatory cell infiltration were significantly reduced,and Mankin's score was significantly decreased.In conclusion,the type Ⅱ collagen can regulate the expression levels of inflammatory factors in the blood of osteoarthritis rats,reduce joint pain and inflammatory cell infiltration,improve synovial tissue inflammation,and promote cartilage hyperplasia,thus re-lieving the symptoms of osteoarthritis.Compared with the positive control,Cartiex™ type Ⅱ collagen has the advantages of less dosage,better effect and faster onset of action.
