培养条件对仙鹤草愈伤组织诱导、增殖及总黄酮含量的影响

以仙鹤草(Agrimonia pilosa Ledeb)无菌苗的子叶为外植体,采用正交设计实验法,研究了不同培养条件对仙鹤草愈伤组织诱导、增殖及总黄酮含量的影响.结果显示:以1/2MS+6-BA 1.0(mg/L)+NAA0.5(mg/L),在pH 6.5时,愈伤组织诱导率较高,且长势良好;以1/2MS+6-BA2.0(mg/L)+NAA 1.0(mg/L),在pH 6.0时,愈伤组织增长最快且总黄酮含量最高.
Effects of Culture Conditions on Callus Induction, Proliferation and the Total Flavonoid Content of Agrimonia pilosa
The cotyledons of Agrimonia pilosa Ledeb were used to study the effect of different culture conditions on callus induction, proliferation and the total flavonoid content through the orthogonal design method. The results showed that: under the condition of 1/2MS + 6 - BA 1.0( mg/L) + NAA 0. 5 (mg/L), at pH 6.5, the callus was induced more easily, and grew well. The condition of 1/2MS + 6 - BA 2.0(mg/L) + NAA 1.0(mg/L) , and pH 6.0 was the most proper condition for callus to proliferate. At the same time, the total flavonoid content in callus was the highest under that condition.
