运用RP-HPLC同时测定火龙果中7种有机酸和Vc含量的方法研究

以紫红肉火龙果为试验材料,对影响有机酸分离的主要因素进行研究,最终建立了运用反相高效液相色谱法(RP-HPLC)同时快速测定火龙果果实中草酸、酒石酸、苹果酸、乙酸、乳酸、柠檬酸、琥珀酸7种有机酸及Vc含量的方法.采用色谱条件为:柱温30 ℃,流速1 mL/min,色谱柱为Kromasil柱(250 mm×4.6 mm),波长215 nm,缓冲液0.05 mol/L H3 PO4-Na2 HPO4;测定结果显示,供试火龙果含有7种有机酸和Vc,苹果酸是其最主要的有机酸、其次是柠檬酸.此方法可以在10 min内完成1次测定,具有分析速度快、灵敏度高、重复性好等优点,为果实中有机酸及Vc的测定及相关代谢研究提供参考.
A Study on Simultaneous Determination of 7 kinds of Organic Acids and Vc in Dragon Fruit by RP-HPLC
The main factors that affect the separation of organic acids were studied,and a method for sim-ultaneous and rapid determination of purple red flesh dragon fruit in oxalic acid,tartaric acid,malic acid,acetic acid,lactic acid,citric acid,succinic acid and Vc content with reverse-phase high performance liquid chroma-tography(RP-HPLC) was established.The chromatographic conditions are as follows:the column temperature was 30 ℃,the flow rate was 1 mL/min,the wavelength was 215 nm,the buffer was 0.05 mol/L H3 PO4-Na2HPO4 and a Kromasil column (250 mm× 4.6 mm) was adopted.The results showed that malic acid was the most important organic acid,the next was citric acid.Because this method can complete a determination within10 min.So it has the advantages of fast,high sensitivity and good repeatability,it is a high reference to the de-termination and research of metabolism of organic acid and Vc in fruit.
