杏鲍菇菌糠漆酶分离纯化及部分酶学性质研究

为寻求制取漆酶廉价的原料,实现杏鲍菇菌糠资源高值化利用,以杏鲍菇菌糠为材料,采用硫酸铵分级沉淀、冷冻干燥和DEAE-sepharose柱层析技术分离纯化漆酶并对其酶学性质进行研究.结果表明:经Native SDS-PAGE检测所得漆酶为单一纯品,分子量大小约为44.3 ku,酶比活6.233 U/mg,纯化倍数10.452,酶活回收率16.640%;该酶氧化ABTS的Km值为0.087 mmol/L,催化氧化底物ABTS的最适反应温度为40℃,低于35℃时有较好的热稳定性,最适pH为3.8,在pH 3.8~4.6表现出较强的稳定性;金属离子Fe2+、Cu2+对漆酶有明显抑制作用,而Fe3+、Mn2+有显著促进作用.
Purification and Properties of Laccase from Spent Mushroom Compost of Pleurotuseryngii
The main aim of this study was to finda cheap raw material,spent mushroom compost of Pleuro-tuseryngii,which can be used to extract laccase.Laccase from mushroom dregsof P.eryngii was graded-precipi-tated with ammonium sulfate,frozen and dried,separated and purified with chromatography on DEAE-cellulose and its enzymatic properties were studied.The resuts showed:the laccase detected by Native SDS-PAGE was a single pure product,its molecular weight was about 44.3 ku,the purification of about 10.452-fold was achieved with an overall yield of 16.640%;the specific activity of the laccase was 6.233 U/mg,the Mc value of its oxi-dation of ABTS was 0.0870 mmol/L;the optimum temperature for laccase activity was 40℃,and the laccase could work continuously under 35℃;the optimum pH for the laccase activity was 3.8,and this laccase showed a good stability when the pH of the buffer varied from 3.8 to 4.6;the laccase activity was enhanced by Fe2+, Cu2+ and was strongly inhibited by Fe3+,Mn2+.
