Effects of Cu2+stress on toxicity and expression of immune related genes in Chinese mitten crab,Eriocheir sinensis
The toxic effects of Cu2+stress on the Chinese mitten crab,Eriocheir Sinensis,were analyzed by using toxicology methods.The results showed that the crabs at the low Cu2+concentration group(0.1 mg/L)and the control group(0 mg/L)had no obvious death,and the crabs at the high Cu2+concentration group(0.5 mg/L and 1.0 mg/L)all died after stress for 72 hours,which indicated that high concentration Cu2+stress had obvi-ous toxic effect on Eriocheir Sinensis.On the basis of this results,the influence of the Cu2+(concentration of 0.1 mg/L)stress on the immune-related genes expression,including lipopolysaccharide and beta-1,3-glucan bind-ing protein(LGBP),Tube,Dorsal,proPO-activating factor(PPAF),glutathione peroxidase(GPx),glutathione-S-transferase(GST),peroxinectin(PXN)was analyzed the using the real-time fluorescent quantitative PCR technique.The data indicated that the LGBP mRNA expression was significantly higher than that of the control group(P<0.05)after stressed for 36 h;the PPAF mRNA expression was significantly higher than that of the control group(P<0.05)after stressed for 12 h and 48 h;the Tube mRNA was significantly higher than that of the control group(P<0.05)after stressed for 48 h,and was significantly lower than that of the control group(P<0.05)after stressed for 72 h;the Dorsal mRNA expression was significantly higher than that of the control group(P<0.05)after stressed for 48 h;the GPx mRNA expression was significantly higher than that of the con-trol group(P<0.05)after stressed for 36 h,and was significantly lower than that of the control group(P<0.05) after stressed for 72 h;the GST mRNA expression was significantly higher than that of the control group (P<0.05)after stressed for 36 h and 48 h;the PXN mRNA expression was significantly higher than that of the control group(P<0.05)after stressed for 12 h,and was significantly lower than that of the control group (P<0.05)after stressed for 72 h.These results show that the Cu2+stress on Eriocheir Sinensis had significant impact on its immune pattern recognition protein genes(LGBP),immune signal transduction related genes (Dorsal,fube)and immune effect related genes(PPAF,GPx,GST,PXN).From what has been discussed a-bove,it can be concluded that heavy metal Cu2+stress has observable toxic effects and has significant impact on the immune related gene expression in the Chinese mitten crab,Eriocheir sinensis.
