Research on queen rearing technology of Apis cerana cerana by cutting honeycomb
[Objective]The Apis cerana cerana is a high-quality native species in China.During the breeding process,the artificial grafting larvae technique has been used to cultivate queens.However,the amount of royal jelly at the bottom of the larvae is relatively small,making it challenging to graft larvae and easy to damage the larvae while raising queens.Therefore,exploring a simple and convenient method of raising queens for the Apis cerana cerana is essential.[Method]A device has been designed to cut honeycomb for queen rearing of Apis cerana cerana,which comprises egg-laying frame,queen-rearing bars,fixing bars,and queen-rearing frames.The device was placed in the bee colony(five honeycombs)to allow worker bees to secrete beeswax for nest construction.A queen was encouraged to lay eggs in it and the frame was placed into the hatching area for eggs incubation.After the hatching of eggs,the queen-rearing bars were dismantled,inserted into the queen-rearing frame,and introduced into a queen-less colony for queen rearing.Parameters such as queen cell length,emergence rate,initial weight of the newly emerged queen,morphological indicators of the queen,development-related genes(Vg,Hex70b,Hex110),and daily egg-laying quantity of the queen were measured.Artificial grafting was used as a control.[Result]The length of the queen cells and the emergence rate of the queens reared through cutting honeycomb group were significantly higher than those in the manual removal group(P<0.05);the initial weight,thorax length,forewing length and forewing width of the queens reared through cutting honeycomb group were significantly higher than those of the queens in the manual group(P<0.05),but the thorax width of the queens reared through cutting honeycomb group was insignificantly different from that of the manual removal group(P>0.05);The number of ovipositor tubes and the daily egg production of queens after natural mating reared through cutting honeycomb group were also significantly higher than that of the manual group(P<0.05).Of the three genes related to queen development(Vg,Hex110,Hex70b),the expression of two genes(Vg,Hex110)in the queens reared through cutting honeycomb group was significantly higher than that of the manual transfer group(P<0.05),and the expression of Hex70b was not significantly different(P>0.05)between the two groups.[Conclusion]The device designed according to the biological characteristics of Apis cerana cerana can effectively cultivate queens,and the quality of queens cultivated by the device is better than that of traditional manual removal.
