Isolation and identification of mulberry fruit sclerotiniosis pathogen and laboratory screening of biological fungicides
[Objective]Mulberry fruit sclerotiniosis is a destructive disease on mulberry fruits,which poses a serious threat to the healthy development of the fruit and mulberry industry and yield stability.In order to provide the theoretical basis for the green prevention and control technology of mulberry fruit sclerotiniosis,the pathogens causing mulberry fruit sclerotiniosis in Le'an County,Jiangxi Province,were isolated and identified and the biopesticides with the ability of inhibiting the growth of the pathogens mycelium were screened.[Method]Tissue isolation method and Koch's rule were used to isolate and determine the pathogenicity of diseased mulberry in Le'an County,Jiangxi Province.Morphological and molecular biology methods were used to identify the isolated pathogens and optimize their cultivation conditions.The inhibitory effect of 7 biopesticides on the pathogen was measured using the drug containing plate method.[Result]Eight strains of fungi were isolated from the sclerotia collected from the planting site where mulberry fruit sclerotiniosis occurred,and cultured on PDA medium with consistent characteristics.The ITS and RPB2 sequences of representative strain JH21 were amplified and sequenced,and the strain JH21 was identified as Scleromitrula shiraiana by homology comparison.The mycelium of S.shiraiana grew better when it was incubated on mung bean dextrose agar medium(MBDA)at pH of 6 for 12 h with alternating light and dark and temperature conditions of 20-25℃.The laboratory screening of fungicides results showed that 0.3%Tetramycin aqueous solution and 0.3%Eugenol soluble solution had good inhibitory effects on the growth of S.shiraiana with EC50 of 0.078 μg/mL and 0.667 μg/mL respectively.[Conclusion]In this study,it was determined that mulberry plantations in Le'an County of Jiangxi Province were mainly threatened by reduced sorosis scleroteniosis caused by S.shiraiana.As biopesticides,Tetramycin and Eugenol are eco-friendly and safe,and they have potential applications in the prevention and control of mulberry fruit sclerotiniosis.
