Construction of Agrobacterium-mediated taro genetic transformation system
[Objective]Taro is a typical clonal crop with narrow genetic background,which makes it difficult to develop new varieties.The construction of a stable genetic transformation system for taro can realize the rapid and accurate improvement of its traits,and promote the transformation of taro from traditional breeding to biological breeding.[Method]Four multi-cormels taro varieties with extensive planting area in China were used as test materials to screen the taro varieties that are easy to induce callus,the stem tips were first exfoliated and cultured on growth medium for 30 days,then transferred to callus induction medium,and further optimize the type and ratio of phytohormones to improve the callus induction rate,proliferation rate and budding rate,based on these results,the genetic transformation efficiency was improved by vacuum treatment and dark treatment.[Result]'GanYu No.2'was selected from four multi-cormels taro varieties to induce callus easily,the callus induction rate in induction medium(MS+sucrose 30 g/L+Agar 6.5 g/L+NAA 0.1 mg/L+TDZ 2.0 mg/L)was 66.7%,the proliferation coefficient in propagation medium(MS+sucrose 30 g/L+Agar 6.5 g/L+NAA 0.1 mg/L+TDZ 2.0 mg/L)was 4.6,the budding rate in bud induction medium(MS+sucrose 30 g/L+Agar 6.5 g/L+NAA 0.5 mg/L+6-BA 1.0 mg/L)was 72.2%.The proportion of positive plants for CeSUC3 was 65.2%and the genetic transformation rate was 5.0%when the callus were vacuum for 15min at-0.1 MPa and cultured in the dark for 7-15 d.[Conclusion]Through variety selection,medium optimization,infection condition and culture condition modified,a stable genetic transformation system of multi-cormels taro mediated by Agrobacterium was established,which laid a foundation for the biological breeding of taro.
