Effects of metabolic and oxidative stress on apoptosis of MCF-7 cells induced by coumaric acid
Objective To investigate the mechanism of apoptosis of breast cancer cells MCF-7 induced by coumaric acid and the effects of oxidative stress and metabolism.Methods The effects of coumaric acid on the oxidative stress of MCF-7 were inves-tigated by the activity of oxidation-related proteins.The effects of coumaric acid on the metabolism of MCF-7 cells were investi-gated by liquid mass spectrometer combined with metabolomics.Results Coumaric acid promoted the apoptosis of MCF-7 cells in a concentration-dependent manner,and up-regulated the oxidative stress of MCF-7 cells,resulting in the imbalance of the oxida-tive system of MCF-7 cells.At the same time,coumaric acid affects the cell metabolism of MCF-7,mainly reflecting glucose metabolism.A total of 563 differential metabolites were identified,of which 463 were up-regulated and 100 down-regulated.Con-clusion The apoptosis of MCF-7 cells induced by coumaric acid may be achieved through the effect of oxidative stress and regu-lation of glucose metabolism.
