Analysis of the mechanism of action of Zuojin pill in the treatment of ulcerative colitis based on MiR-21
Objective By observing the effect of Zuojin Pill on ulcerative colitis(UC)model mice,the mechanism of action of Zuojin Pill in the treatment of ulcerative colitis was studied from the miR-21→STAT3/PI3K signaling pathway.Methods Sixty BALB/c mice were randomly divided into a blank group,a model group,a mesalazine group(MS,0.3 g/kg),and a Zuojin pill group(6 g/kg),and an acute UC mouse model was constructed with dextran sodium sulfate(DSS),and the drug was administered by gav-age starting on the second day of modelling and continuing for 7 days,and the general conditions of the mice were observed.HE staining was used to observe the morphology of mouse colon tissue,and Western blot method was used to detect the expression of STAT3,PI3K and AKt proteins in mouse colon tissue.Quantitative Real-time PCR was used to detect miR-21 gene expression in mouse colon tissue.Results Zuojin pill solution and MS solution had significant therapeutic effects on UC model mice,and the ef-fect was more significant in the Zuojin pill group of mice,reducing the damage to colon tissue by inhibiting STAT3,PI3K,AKt and miR-21.Conclusion Zuojin pill is effective in immunomodulation of UC mice,and miR-21 should be the key link of Zuojin pill intervention in UC mice.
