Study on the Mechanism of NR4A1 Regulating Apoptosis Process and Oxidative Stress Injury in PD Cell Model
Objective To analyze the mechanism of NR4A1,a member of orphan nuclear receptor superfamily,regulating the apoptosis process of Parkinson's disease(PD)cell model and oxidative stress injury.Methods Through SH-SY5Y cell culture,transfection and MMP+treatment,PD cell model was established,and NR4A1 overexpression vector,empty vector and NR4A1 specific silencing vector were transfected into the cell model,and the activities of NR4A1,caspase-3,Bcl-2 and Bax protein levels in different groups of cell models were measured,and the activities of lactate dehydrogenase(LDH)and reactive oxygen species(ROS)in different groups of down-regulated models were analyzed.Results The expression level of NR4A1 mRNA and protein in NR4A1 overexpression group was higher than that in NR4A1 empty vector group,and the expression level in NR4A1 empty vector group was higher than that in NR4A1 silent group,with statistical significance(P<0.05).The levels of Bax protein,caspase-3 activ-ity,LDH,ROS and SOD in NR4A1 overexpression group were lower than those in NR4A1 empty vector group,and the levels of Bax protein,Caspase-3 activity,LDH,ROS and SOD in NR4A1 empty vector group were lower than those in NR4A1 silent group,with statistical significance(P<0.05).Conclusion NR4A1 can affect the apoptosis process of PD cell model.Overexpression of NR4A1 can help to inhibit the apoptosis of PD cells,and also has a positive effect on alleviating the oxidative stress damage of PD cells.
