Therapeutic mechanism of solanine in osteosarcoma 143B cells
Objective To investigate the effects of solanine on proliferation,cloning,migration and invasion of osteosarcoma 143B cells,and explore the mechanism of solanine in the treatment of osteosarcoma.Methods 143B cells of osteosarcoma were treated with solanine at different concentrations(0,20,40,80,160 μg/mL).The effect of solanine on the proliferation of 143B cells was detected by CCK-8 assay.143B cells were treated with solanine at concentrations of 0,40 and 80 μg/mL.The effects of sola-nine on the cloning,migration and invasion ability of 143B cells were detected by cloning test and Transwell test.The expressions of apoptosis related proteins Bax and Bcl-2 and autophagy related proteins P62,Beclin-1 and LC3 were detected by Western blot-ting test.Results Compared with the control group,solanine could significantly inhibit the proliferation,cloning,migration and in-vasion of 143B cells in a dose dependent manner(P<0.05).After solanine treatment,the expression of apoptosis-related protein Bax increased,the expression of Bcl-2 decreased(P<0.05),the expression of autophagy related protein P62 decreased(P<0.05),the expression of Beclin-1 protein and the ratio of LC3-Ⅱ/LC3-Ⅰincreased(P<0.05).Conclusion This study shows that sola-nine can inhibit the proliferation,invasion and migration of osteosarcoma 143B cells.It can also play the role of anti-osteosarcoma 143B cells by inducing apoptosis and promoting autophagy.
