评估茶枝柑黄龙病病情的3种定量方法的比较研究

[目的]为比较3种不同黄龙病病情定量评估方法的优劣之处。[方法]运用实时荧光定量PCR(qRT-PCR)技术,定量分析了柑橘黄龙病菌(Candidatus Liberibacter asiaticus,CLas)在茶枝柑的叶脉、叶肉、果皮、囊壁、果实中柱、树皮、根系等多个部位的分布情况,并进一步使用"橘园黄龙病病情指数分析、病树中CLas 含量分析及黄龙病发病率检测"3 种方法定量评估了新会区两个茶枝柑果园中黄龙病的发病情况。[结果]CLas在茶枝柑各部位含量的大小:叶脉>橘络>果实中柱>根>叶肉>绿树皮>果皮白瓤>囊壁>外果皮。茶枝柑叶脉中CLas 16S rDNA基因平均含量为107。89 CN/g FW,茶枝柑外果皮中CLas 16S rDNA基因平均含量为104。54 CN/g FW。三江镇联合村果园 20棵茶枝柑的平均病情指数为 52。75;随机采集叶片样品,可在其中18 棵茶枝柑的叶脉样品中检出CLas,其 16S rDNA平均含量为 105。57 CN/g FW;选择性采集显症叶片样品,可检测出 20 棵茶枝柑树的黄龙病发病率为 100%。双水镇大树村果园 20 棵茶枝柑的平均病情指数为 30。75;随机采集叶片样品,可在其中 3 棵茶枝柑的叶脉样品中检出CLas,其 16S rDNA基因的平均含量为 104。99 CN/g FW;选择性采集显症叶片样品,可检测出 20 棵茶枝树的黄龙病发病率为 100%。[结论]明确CLas在茶枝柑不同部位的定量分布情况,为茶枝柑的黄龙病定量分析提供了参考依据;3 种定量评估茶枝柑黄龙病病情的方法各有优劣,综合运用才能更准确全面地反映出黄龙病在果园的发病情况。
A comparative study of 3 quantitative methods for evaluating the damage of Citrus Huanglongbing in Citrus reticulata cv.'Chachiensis'
[Objective]This study was conducted to compare the advantages and disadvantages of three different quatitative assessment methods for Huanglongbing.[Method]To fully understand the damage of Huanglongbing(HLB)to Citrus reticulata cv.'Chachiensis',the quantitative real-time PCR(qRT-PCR)was used to quantitatively analyze the distribution of Candidatus liberibacter asiaticus(CLas)in the veins,mesophyll,peel,sac wall,stele,bark and root of C.reticulata cv.'Chachiensis'.The incidence of HLB damage in two orchards of C.reticulata cv.'Chachiensis'were quantitatively evaluated by three methods:the disease index analysis of HLB,CLas content analysis in diseased trees and detection of HLB incidence.[Result]The results showed that the CLas contents in different parts of C.reticulata cv.'Chachiensis'tree were as follows:vein>tangerine meridians>fruit stele>root>mesophyll>green bark>peel white pulp>sac wall>exocarp.The content of the 16S rDNA gene of CLas in the veins of C.reticulata cv.'Chachiensis'was 107.89 CN/g FW,and the content of the 16S rDNA gene of CLas in the exocarp of C.reticulata cv.'Chachiensis'was 104.54 CN/g FW.The average disease index of 20 C.reticulata cv.'Chachiensis'trees in the orchard of Lianhe Village in Sanjiang Town was 53;the leaf samples were collected randomly,and CLas was detected in the vein samples of 18 C.reticulata cv.'Chachiensis'trees,with an average content of 105.57 CN/g FW;by selective collection of symptomatic leaf samples,the incidence of HLB in 20 C.reticulata cv.'Chachiensis'trees was 100%.The average disease index of 20 C.reticulata cv.'Chachiensis'trees in Dashu Village orchard of Shuangshui Town was 32.CLas was detected in the vein samples of three C.reticulata cv.'Chachiensis'trees,and the average content of 16S rDNA gene was 104.99 CN/g FW.By selectively collecting symptomatic leaf samples,the incidence of HLB in 20 C.reticulata cv.'Chachiensis'trees was 100%.[Conclusion]This study investigated the quantitative distribution of CLas in different parts of C.reticulata cv.'Chachiensis'trees,and provided a reference for the quantitative analysis of HLB in C.reticulata cv.'Chachiensis'.At the same time,this study also showed that three methods of HLB damage assessment had their own advantages and disadvantages,and the comprehensive application of three methods could more accurately reflect the incidence of HLB in the orchard.
