Analysis of the Genetic Diversity of Alternaria alternata in Jiangxi Province
[Objective]To clarify the genetic diversity of Alternaria alternata in Jiangxi Province,relevant studies were conducted.[Method]Random Amplified Polymorphic DNA(RAPD)molecular markers were used to analyze 37 isolates of Alternaria alternata from 6 major tobacco-producing areas in Jiangxi Province.[Result]Nei's gene diversity index(He)of 6 different geographical populations was 0.161 8~0.320 9,and Shannon information index(I)was 0.241 9~0.478 5.A.alternata from different geographical sources had rich genetic diversity,and genetic distance analysis showed that strains from Xingguo and Xinfeng counties had the smallest genetic distance and close genetic relationship,while that form Shicheng and Lichuan had the largest genetic distance and far genetic relationship.The migration number per generation(Nm)of the population was 1.817 1,indicating that there was a certain degree of gene flow among populations from different geographical origins.Using NTSYS software and UPGMA method for cluster analysis,when the similarity coefficient was defined as 0.56,37 strains could be divided into 10 groups,and 37 strains were basically clustered into the corresponding branches by geographical origin,indicating that there was obvious correlation between genetic diversity among strains and geographical origin.[Conclusion]The genetic diversity of A.alternate isolated from 6 tobacco-producing areas in Jiangxi Province was confirmed by RAPD,making the basis for the tobacco varieties against A.alternate as well as the disease control.
