Identification of the pathogen of Smilax china in Yizhang County of Hunan Province
[Objective]In November 2023,a leaf rust disease was discovered on Smilax china in Yizhang County,Hunan Province.To investigate the cause of the disease,relevant researches were conducted.[Method]The morphology of the diseased part of the leaves was observed under the optical microscope,and then the total DNA of the diseased part of the leaves was extracted,and the ITS and LSU sequences of pathogen were amplified with universal primers.The amplified fragments were recovered by cutting the gel,and sequenced.The sequencing results were compared with the NCBI database and phylogenetic tree was conducted.[Result]The pathogen of rust fungus on Smilax china was identified as Puccinia sp..[Significance]The study provided references for the prevention and control of the disease in the future.
