Mutation Breeding of High-yield Protease Strains and Evaluation of Its Feather-degrading Ability
This study aimed to screen protease-producing soil bacterial resources,bacterial isolation culture and enrichment culture methods were used to separate protease-producing bacteria in the soil of the feather stacking area of chicken farms.Based on this strain,the strain with high protease activity was selected by ultraviolet mutation and its ability to degrade feathers was evaluated.The results showed that after the initial screening,rescreening and mutagenesis,the protease activity of the mutagenic strain increased from 31.33 U/mL to 42.13 U/mL,and the enzyme activity increased by 34.47%;after 7 days of cultured in the medium containing chicken feathers,the feather degradation rate of the screened starting strain was 51.3%,while the feather degradation rate of the mutagenic strain was as high as 80.6%,which was significantly higher than that of the starting strain,providing an important prerequisite and foundation for the development of excellent protease-producing microecological preparations.
