高效液相色谱法测定天然彩色茧中叶黄素

摘要
本研究采用超声波萃取,高效液相色谱法测定天然彩色茧中叶黄素,色谱柱为ZORBAZ Extend C18(4.6×150 mm,5 μm),流动相为甲醇∶水(95∶5),流速为 1 mL/min,检测波长为 450 nm.结果显示,叶黄素浓度在 0.5 μg/mL～10 μg/mL范围内呈线性,相关系数R2=0.999,方法回收率为 96%～100%,相对标准偏差为 0.33%,表明该方法可用于天然彩色茧中叶黄素的测定.
Abstract
The lutein in natural color cocoons was extracted by ultrasonic and detected by high performance liquid chromatography,the chromatography was performed on ZORBAZ Extend C18(4.6×150 mm,5 μm)with mobile phase consisting of methanol∶water(95∶5),the flow rate was 1 mL/min,the detection wavelength was 450 nm.The results showed that the lutein concentration exhibits linearity within the range of 0.5 μg/mL to 10 μg/mL,with a correlation coefficient R2 of 0.999,the recovery was 96%-100%,and the relative standard deviation was 0.33%,it is thus clear that the method could be used for the determination of lutein in natural color cocoons.
