GB 4789.15-2016第一法与旧版相比,最大的改动就是由倒置培养法变为正置培养法,但正置培养法有时会出现空白对照被污染的情况.本文从"人、机、料、法、环"5要素设计试验,发现空白对照被污染主要是由于培养箱内的鼓风系统引起的空气流动所致.通过风速增加试验,确证其原因,发现通过外加透气保护袋试验,可以解决正置培养法污染问题.
Analysis of the reasons for contamination in the upright cultivation method about GB 4789.15-2016 and corresponding solutions
Compared with the previous edition,the most significant alteration in the first method of GB 4789.15-2016 is the shift from the inverted culture method to the upright culture method,but the upright culture method sometimes leads to the contamination of the blank control.In this paper,experiments were designed based on the five elements of"human,machine,material,method and environment",found that the contamination of the blank control is mainly attributed to the air flow caused by the blower system inside the incubator,and through the wind speed increase test,the cause was confirmed.The contamination issue of the upright culture method can be resolved through the addition of a breathable protective bag test.
