Objective To investigate the predictive value of microRNA-132 and its target gene NAG-1 in evaluating the therapeu-tic effect of nonsteroidal anti-inflammatory drugs in patients with ankylosing spondylitis(AS).Methods 154 patients with AS in oui hospital were selected as the observation group,and 154 healthy subjects who underwent physical examination in our hospital in the same period were selected as the control group.The expression levels of miR-132 and NAG-1 mRNA in the two groups were observed.Patients in the observation group were treated with diclofenac sodium.The expression levels of miR-132 and NAG-1 mRNA before treatment were observed to predict the efficacy of American Rheumatology Association(ACR)20 after treatment.Results The ex-pression level of miR-132 mRNA in mononuclear of patients in the observation group was higher than that in the control group,and the expression level of NAG-1 mRNA was lower than that in the control group(P<0.05).The sensitivity of miR-132 in predicting AS was 83.8%,the specificity was 98.1%,the best cut-off value was 1.373,the area under the curve(AUC)was 0.964,and the 95%CI was 0.936-0.982.The sensitivity of NAG-1 in predicting AS was 76.0%,the specificity was 86.4%,the best cut-off value was 0.826,AUC was 0.879,and 95%CI was 0.838-0.914.The expression level of miR-132 mRNA in ACR20 group was lower than that in non ACR20 group,and the expression level of NAG-1 mRNA was higher than that in non ACR20 group(P<0.05).The sensitivity of miR-132 in predicting ACR20 was 81.8%,the specificity was 94.2%,the AUC was 0.950,and the 95%CI was 0.903-0.979.The sensitivity of NAG-1 to predict ACR20 was 97,0%,the specificity was 46.3%,the AUC was 0.751,and the 95%CI was 0.675-0.817.Conclusions miR-132 and NAG-1 may be potential biological indicators of the therapeutic effect of AS and non steroidal anti-inflammatory drugs.
