Effect of Sphingosine-1-phosphate Receptor Agonist Fingolimod on the Proliferation and Migration of Melanoma B16-F10 and Its Mechanism
Objective To observe the effects of fingolimod on proliferation and autophagy-related pathway proteins in melanoma.Methods B16-F10 cells were cultured in DMEM medium containing different concentrations of fingomode(0,5,10 and 15 μM)for 24 h,and cell viability was detected by CCK-8 cell viability assay kit;the migration ability of B16-F10 cells was detected by scratch assay;trypan blue staining was used to detect cell death;immunofluorescence assay and Western Blot assay were used to de-tect the expression of autophagy related pathway proteins.Results Fengomode significantly inhibited the viability and proliferation of B16-F10 cells in a dose-dependent manner;fengomode weakens the migration ability of B16-F10 cells and causes cell death;immu-nofluorescence and Western Blot showed that compared with the control group,the expressions of autophagy pathway-related proteins LC3,Beclin1 and P62 in FTY-720 group were changed(P<0.05).Conclusion Fengomode can inhibit the proliferation of melano-ma B16-F10 cells and cause cell damage,and the mechanism may be related to the autophagy of B16-F10 cells induced by fengo-mode.
