Berberine Inhibits Inflammation via the JAK2/STAT3 Pathway to Repair Spinal Cord Injury
Objective To observe the effect of berberine on the recovery of nerve function in mice with spinal cord injury and ex-plore its mechanism.Methods The mouse model of spinal cord injury was prepared,and the hind limb motor function was detected by BMS score;the number of neurons in spinal cord was observed by NeuN immunofluorescence staining;the contents of IL-6,TNF-α,IL-1β and IL-10 were detected by ELISA;the mRNA expression of TNF-α,IL-6,TGF-β1 and Arg-1 was detected by qRT-PCR;P-JAK2,JAK2,P-STAT3 and STAT3 proteins were detected by Western Blot.Results Berberine improves motor function in mice with spinal cord injury,increases the number of neurons,decreases the secretion of pro-inflammatory factors TNF-α,IL-1β and IL-6,and increases the secretion of anti-inflammatory factor IL-10.Berberine decreases the expression of TNF-α mRNA and IL-6 mRNA,and increases the expression of Arg-1 mRNA and TGF-β1 mRNA.Conclusion berberine reduces inflammation,in-creases neuronal survival,and promotes motor function recovery in mice with spinal cord injury through JAK2/STAT3 signaling path-way.
