Study on the Effect and Mechanism of Roxburghil Polysaccharide Combined with Palbocinb in the Treatment of Pancreatic Cancer
Objective To investigate whether Roxburgh polysaccharide promotes the efficacy of Palbocinb in the treatment of pan-creatic cancer by targeting the elimination of senescent tumor cells.Methods Firstly,the proliferation rate of PANC02 cells treated with Palbocinib was detected by CCK8 method,and the expression of Ki-67 and PCNA genes in PANC02 cells was detected by real-time quantitative PCR(qRT-PCR)method;then SA-β-Gal staining kit and SA-β-Gal staining fluorescent probe were used to de-tect the proportion of SA-β-Gal expression in PANC02 cells;In vitro,the effects of Palbocinib on PANC02 cell proliferation were de-tected by CCK8 method,and the apoptosis of PANC02 cells was detected by flow cytometry;at the same time,subcutaneous tumor bearing model was used for in vivo experiment.The mice were given Roxburghil polysaccharide and Palbocinb by gavage,the tumor volume was measured,the tumor mass was weighed,and the survival period and recurrence rate were recorded;finally,SA-β-Gal staining fluorescence probe was used to detect the proportion of SA-β-Gal expression in mouse tumor cells in each group.Results Palbocinb could effectively inhibit the proliferation of PANC02 cells and decrease the expression of Ki-67 and PCNA in PANC02 cells at the gene level.At the same time,Palbocinib induced senescence of PANC02 cells and increased the expression of SA-β-Gal.Compared with the single treatment group,Palbocinib combined with Roxanthus polysaccharide could inhibit the proliferation and in-crease the apoptosis of PANC02 cells more effectively in vitro.After in vivo treatment,the tumor mass and volume decreased signifi-cantly,the survival period was prolonged,and the recurrence rate was reduced.The proportion of senescent cells in mouse tumor cells was decreased effectively by roxburghii polysaccharide.Conclusion Roxanthus polysaccharide can effectively synergistically in-hibit the growth of pancreatic cancer cells in vitro and in vivo,possibly through targeted elimination of Palbocinib induced senescence of pancreatic cancer cells.
