Expression and Clinical Significance of β-1,3-galactosyltransferase-1 and β-1,3-galactosyltransferase-2 in Breast Cancer

目的 检测β-1,3 半乳糖基转移酶-1(β-1,3-galactosyltransferase-1,B3GALT1)和β-1,3 半乳糖基转移酶-2(β-1,3-galactosyltransferase-1,B3GALT2)在乳腺癌组织及癌旁正常组织中的表达及其与临床病理特征之间的关系,并分析其相关通路.方法 选取乳腺癌及癌旁组织标本 132 例,采用 Western Blot 法和免疫组化方法检测 B3GALT1 和B3GALT2 在乳腺癌及癌旁正常组织中的表达,分析其临床意义.通过基因集富集分析法探索相关通路.结果 乳腺癌组织中B3GALT1 和B3GALT2 表达均低于癌旁正常组织(P<0.05),二者表达呈正相关(r=0.635,P<0.001),且两者表达阳性患者与阴性患者的T分期和Ki-67 比较,差异均具有统计学意义(P<0.05).B3GALT1 和B3GALT2 均富集于JAK-STAT通路和Notch通路.结论 B3GALT1 和B3GALT2 在乳腺癌中低表达,与乳腺癌的增殖、生长密切相关.
Objective To investigate the expression and their relationship with clinical pathological features of β-1,3-galactosyl-transferase-1(B3GALT1)and β-1,3-galactosyltransferase-2(B3GALT2)in breast cancer,and analyze their related signaling path-ways.Methods The expressions of B3GALT1 and B3GALT2 in breast cancer and adjacent normal tissues were detected by Western Blot and immunohistochemical methods in 132 cases of breast cancer and adjacent tissues,and their clinical significance was analyzed.The related pathways were explored by gene set enrichment analysis.Results The expressions of B3GALT1 and B3GALT2 in breast cancer tissues were lower than those in adjacent normal tissues(P<0.05),and the expressions of the two were positively correlated(r= 0.635,P<0.001),and the differences in T stage and Ki-67 between positive and negative patients were statistically significant(P<0.05).Both B3GALT1 and B3GALT2 are enriched in JAK-STAT pathway and Notch pathway.Conclusion B3GALT1 and B3GALT2 are significantly downregulated in breast cancer and closely related to the proliferation and growth of breast cancer.
