甘草附子汤调控巨噬细胞极化治疗痛风性关节炎

目的 本研究旨在探究甘草附子汤对由尿酸钠晶体引发的大鼠痛风性关节炎的影响,并进一步研究其通过调节巨噬细胞极化途径的潜在机制.方法 本实验共纳入40 只SD大鼠,随机分配成对照组、模型组、秋水仙碱组和甘草附子汤组.利用MSU注射入大鼠的踝关节腔内,构建痛风性关节炎的动物模型.通过大鼠足部肿胀情况及其步态变化来评估症状严重程度;使用HE染色技术对关节组织样本进行组织学评分;通过酶联免疫吸附试验来测定大鼠血浆IL-1β、IL-10 含量;采用Real-Time PCR法测定大鼠关节中iNOS、Arg-1 的表达;采用免疫荧光测定TNF-α在滑膜组织的表达.结果 甘草附子汤能明显减轻痛风性关节炎大鼠足跖肿胀,改善步态及组织学评分(P<0.05).大鼠经过甘草附子汤的治疗,血清中IL-1β表达水平显著降低(P<0.05),相反IL-10 表达水平显著升高,其中P<0.05;Real-Time PCR结果提示,甘草附子汤能显著降低iNOS的表达(P<0.01),增加Arg-1 表达(P<0.01).同时免疫荧光检测发现甘草附子汤抑制TNF-α的表达.结论 甘草附子汤能有效抑制痛风性关节炎的发展,其作用机制可能与促进巨噬细胞从M1 型向M2 型的转变密切相关.
Treatment for Gouty Arthritis by Regulating Macrophage Polarization with Gancao Fuzi Decoction
Objective To investigate the effect of Gancao Fuzi Decoction on gouty arthritis induced by sodium urate crystals in rats,and to further investigate the potential mechanism of Gancao Fuzi Decoction by regulating the polarization pathway of macropha-ges.Methods A total of 40 SD rats were included in this experiment and randomly assigned into control group,model group,colchi-cine group and Gancao Fuzi Decoction group.MSU was injected into the ankle cavity of rats to construct an animal model of gouty ar-thritis.The severity of symptoms was assessed by foot swelling and gait changes in rats;HE staining technique was used to score the joint tissue samples;plasma IL-1β and IL-10 were determined by enzyme-linked immunosorbent assay(ELISA);the expression of iNOS and Arg-1 in the joints of rats was determined by Real-Time PCR;the expression of TNF-α in synovial tissue was determined by immunofluorescence.Results Gancao Fuzi Decoction can significantly reduce the swelling of foot and plantar of rats with gouty ar-thritis,and improve gait and histological score(P<0.05).After treatment with Gancao Fuzi Decoction,serum IL-1β expression lev-el was significantly decreased(P<0.05),the expression level of IL-10,however,was significantly increased(P<0.05);at the same time,Real-Time PCR results showed that the expression of iNOS was significantly decreased(P<0.01)and the expression of Arg-1 was increased(P<0.01);meanwhile,immunofluorescence detection showed that Gancao Decoction inhibited the expression of TNF-α.Conclusion Gancao Fuzi Decoction demonstrates a pronounced suppressive impact on gouty arthritis.This effect is likely associated with its role in facilitating the transition of macrophages from the M1 to the M2 phenotype.
