Study of miR-613 Blocking Progression of Cervical Cancer and Its Mechanism
Objective To investigate the effect and mechanism of miR-613 on the progression of cervical cancer cells.Methods HeLa cells were randomly divided into control group,NC group and miR-613 overexpression group.The expression of miR-613 in HeLa cells and E6E7 cells was detected by real-time fluorescence quantitative polymerase chain reaction(RT-qPCR);the prolifera-tion of HeLa cells was detected by Cell Counting Kit(CCK-8);the number of clone cells was detected by clone formation assay.The cells migration was detected by scratch assay;the cells invasion was detected by Transwell assay.The relative expression levels of β-catenin,vimentin,E-cadherin,c-Myc gene and matrix metalloproteinase 9(MMP-9)were detected by Western Blot.Results The relative expression level of miR-613 in HeLa cells was significantly lower than that in normal cervical cells(P<0.05).There was no significant difference in OD value(450 nm),number of clone cells,number of migration and invasion cells and relative expres-sion levels of β-catenin,vimentin,E-cadherin,c-Myc and MMP9 between control group and NC group at any time point(P>0.05);compared with control group and NC group,OD value(450 nm),number of clone cells,number of migration and invasion cells and relative expression levels of β-catenin,vimentin,c-Myc and MMP9 were significantly decreased in miR-613 overexpres-sion group,while relative expression level of E-cadherin was significantly increased in miR-613 overexpression group(P<0.05).Conclusion The overexpression of miR-613 can inhibit malignant biological behaviors(proliferation,migration,invasion)of cer-vical cancer,and thus block the progression of cervical cancer,which is related to miR-613 inhibiting Wnt/β-catenin signaling path-way related proteins.
