The Effects of MST1R Inhibitor BMS-777607 on Propagation and Apoptosis of Ovarian Cancer SKOV3 Cells
Objective To investigate the application of MST1R inhibitor BMS-777607 in ovarian cancer SKOV3 cells and ob-serve its effects on proliferation and apoptosis,and clarify its mechanism of action.Methods The experiment was divided into a con-trol group(DMSO)and an experimental group(BMS-777607)based on whether drug was added to ovarian cancer SKOV3 cells.Different concentrations of BMS-777607 were used to treat ovarian cancer SKOV3 cells.MTT assay,clone formation assay,and EdU cell imaging were used to detect cell proliferation rate.Flow cytometry was used to detect the rate of apoptosis,and Western Blot was used to detect the protein expression level of Bax and p-ERK.Results In the MTT experiment,compared with the control group,the proliferation rate of SKOV3 cells in the treated group significantly decreased with increasing drug concentration(P<0.05).Flow cytometry experiment showed a significant increase in the proportion of SKOV3 cell apoptosis in the treated group(P<0.05).Western Blot experiment showed that compared with the control group,the expression level of Bax protein significantly increased(P<0.01)while the expression level of p-ERK protein significantly decreased(P<0.05)in the SKOV3 cells of the treated group as the drug concentration increased.Conclusion The MST1R inhibitor BMS-777607 can inhibit the proliferation and induce apoptosis of SKOV3 cells.
